Looking for pathways related to COVID-19: confirmation of pathogenic mechanisms by SARS-CoV-2–host interactome

Abstract In the last months, many studies have clearly described several mechanisms of SARS-CoV-2 infection at cell and tissue level, but interaction between host SARS-CoV-2, determining grade COVID-19 severity, are still unknown. We provide a network analysis on protein–protein interactions (PPI) viral proteins to better identify biological responses, induced by both whole proteome specific proteins. A host-virus interactome was inferred, applying an explorative algorithm (Random Walk with Restart, RWR) triggered 28 SARS-CoV-2. The PPI allowed estimate distribution in cell. Interactome built around one single protein define different response, underlining as ORF8 ORF3a modulated cardiovascular diseases pro-inflammatory pathways, respectively. Finally, network-based approach highlighted possible direct action NS7b enhancing Bradykinin Storm. This representation could be framework for pathogenic evaluation clinical outcomes. identified responses important role accessory phenotypes severe patients.
